چکیده

Extensive evidence supports an important role for soluble oligomers of the amyloid β-protein (Aβ) in Alzheimer's Disease pathogenesis. In the present study we combined intracerebroventricular (icv) injections with brain microdialysis technology in the fully conscious rat to assess the effects of icv administered SDS-stable low-n Aβ oligomers (principally dimers and trimers) on excitatory and inhibitory amino acid transmission in the ipsilateral dorsal hippocampus. Microdialysis was employed to assess the effect of icv administration of Aβ monomers and Aβ oligomers on dialysate glutamate, aspartate and GABA levels in the dorsal hippocampus. Administration of Aβ oligomers was associated with a +183% increase (p.

Zataria multiflora could improve hippocampal tau protein and TNFα levels and cognitive behavior defects in a rat model of Alzheimer's disease

Objective: Zataria multiflora (ZM) is a plant with ethnopharmacological value which was recently tested to reduce symptoms of Alzheimer’s disease (AD). The aim of the present study was to determine the effect of ZM essential oil on spatial cognitive and noncognitive behavior, as well as hippocampal tau protein and tumor necrosis factor alpha (TNFα) concentrations in ra...

The beneficial effects of riluzole on GFAP and iNOS expression in intrahippocampal Aβ rat model of Alzheimer’s disease

Background and Objective: Alzheimer’s disease (AD) is a neurodegenerative disorder specified by deposition of b-amyloid (Ab) and neuronal loss that leads to learning and memory disturbances. One of the most important causes of AD is glutamate-dependent excitotoxicity in brain regions that is vulnerable to AD. According to previous reported results, it was revealed that riluzole, as a glutamate ...

Berberine attenuates convulsing behavior and extracellular glutamate and aspartate changes in 4-aminopyridine treated rats

Objective(s): K+ channel blocker 4-aminopyridine (4-AP) stimulates the release of glutamate from nerve terminals and induces seizures. Berberine as a potential herbal drug exerts several pharmacological actions on the central nervous system including anxiolytic, anticonvulsant, and neuroprotective properties. The present study aimed to investigate the effect of berberine on seizure onset and ti...

Effect of Chronic Intracerebroventricluar Administration of Lipopolysaccharide on Connexin43 Protein Expression in Rat Hippocampus

Background: Hippocampal damages, which are accompanied by inflammation, are among the main causes of epilepsy acquisition. We previously reported that chronic intracerebroventricular (i.c.v.) injection of lipopolysaccharide (LPS) modulates epileptogenesis in rats. There is a network of gap junction channels in the hippocampus that contribute to epileptogenesis. Gap junction channels are formed ...

Expression of the type 1 metalloproteinase in the rat hippocampus after the intracerebroventricular injection of β-amyloid peptide (25-35).

The expression of matrix metalloproteinase of the first type was studied in frontal sections of the adult rat brain one month after a single intracerebroventricular injection of beta-amyloid peptide (25-35), which is known to be a well-known model of the development of Alzheimer's disease. Brain sections were stained immunocytochemically to detect MMP-1 expression, and histologically to reveal ...
